Abstract
Shoot tip explants of Friederick's Dendrobium Orchid (Dendrobium friedericksianum Rchb.f) were cultured on solidified Murashing and Skoog (MS) medium supplemented with various types of sugar, agar and various concentrations of 2,4-dichorophenoxyacetic acid (2,4-D) and thidiazuron (TDZ). The pH of medium was adjusted to 5.7 before autoclaving. The medium supplemented with 1 mg/l 2,4-D and 2 mg/lTDZ provided the highest percentage of protocorm-like bodies (PLBs) (51%), and PLB-produced shoots (PLB-PS) (3%). The combination of 1 mg/l 2,4-D and 1.5 mg/l TDZ gave 47% PLB formation and 2% PLBPS. A 3% sucrose resulted in the highest number of PLBs at 51.21%. Phytagel at concentration of 0.17% gave the highest percentage of PLBs (51.65% )
